WASHINGTON, D.C. April 4, 2013 A young scientist's astute observation that grasshoppers don't get sick eventually led to profound insights into our own immunity. Jules Hoffmann, PhD, Professor of Integrative Biology at Strasbourg University, Institute of Advanced Science, France Emeritus, and 2011 Nobel Prize recipient in Physiology or Medicine is the scientist who made these insights. He described his scientific journey in the keynote lecture on April 3, 2013, the opening night of the Genetics Society of America's 54th Annual Drosophila Research Conference in Washington, D.C., April 3-7, 2013.

A father who was "an enthusiastic entomologist" and a high school biology teacher sparked Dr. Hoffmann's early interest in insects, in post-World War II Luxembourg. Later, Dr. Hoffmann marveled at the apparent ability of grasshoppers to evade infection, even when he transplanted organs among them as an undergraduate at the University of Strasbourg. How did this happen?

For his doctoral work at the Institute of Zoology in Strasbourg, France, he discovered a blood-producing tissue near the insect's heart that when damaged with X-rays, led to body-wide infection and failure to molt. After earning his PhD in experimental biology in 1969, Dr. Hoffmann pursued endocrinology for postdoctoral work. In 1978 he became director of the laboratory of endocrinology and immunology at the University of Strasbourg.

In 1990, Dr. Hoffmann's lab replaced the grasshopper with the fruit fly, Drosophila melanogaster, and moved on to a genetic dissection of antimicrobial defense. The group focused on innate immunity, the ancient arm of the immune defense that provides generalized protection against common pathogens.

"At that time, innate immunity was not considered as essential and hardly studied at all. Immunity was mainly seen in the context of antibodies and vaccination" the responses of the adaptive (acquired) immune response, he said. Innate immunity is front-line and immediate; adaptive immunity has memory and specificity.

Dr. Hoffmann and his colleagues' expected the innate immune system in the fly to be completely different from that in a mammal, but they were in for a surprise. Within a few years, thanks to Dr. Hoffmann's lab and the work of several other labs in mammals, including those of Charles A. Janeway, MD, and Bruce A. Beutler, MD, they showed that flies and mammals share many of the mechanisms of innate immunity. "These range from receptors for microbial aggressors, to intracellular signaling cascades, which control the expression of immune response genes, and the genes encoding potent antimicrobial peptides," Dr. Hoffmann explained.

Genomic analysis enabled the Hoffmann group to set the date for the emergence of innate immunity. "Innate immunity appeared with multicellularity, possibly one billion years ago. The various zoological phyla have played with a toolbox of genes encoding receptors, adaptors, kinases, transactivators, and antimicrobial peptides according to their own agendas, essentially their own environments and pathogens," he said.

At the crux of the Hoffmann lab body of work is the Toll receptor, a protein that straddles the cell membranes of immune system "sentinel" cells, detecting molecular nametags of pathogens from the outside and transducing messages to the inside, activating and orchestrating defense against infection.

The Toll receptor had been discovered in 1985 as a regulator of embryo polarity, but Dr. Hoffmann's group linked it to fungal infection in flies in 1996 and then to Gram-positive bacterial infections. During this time Dr. Hoffmann became director of the CNRS Institute of Cellular and Molecular Biology. A year later, Dr. Janeway and his lab at Yale discovered Toll-like receptors that activate and amplify the specific, adaptive immune response in humans.

Dr. Hoffmann acknowledged many scientists and labs who helped him in the discovery that Toll was the sensor of innate immunity in the fly. In 2011 he received the Nobel Prize in Physiology or Medicine shared with Dr. Beutler, who had shown that Toll-like receptors performed a similar role in mammals, and Ralph M. Steinman, MD, for his discovery of the dendritic cell and its roll in adaptive immunity.

As if a foreshadowing of the year to come, in 2010, Dr. Hoffmann received the Keio Medical Science Prize with Shizuo Akira, MD, PhD. But 2011, turned out to be the year of celebration for Dr. Hoffmann. Not only did he receive the Nobel Prize with Drs. Beutler and Steinman, but he also received the CNRS Gold Medal, the Shaw Prize in Life Science and Medicine with Drs. Beutler and Ruslan M. Medzhitov, and the Canada Gairdner Award for Medical Research with Dr. Akira.

But the broader lesson from Dr. Hoffmann's work may be its celebration of basic research. "Our work on antimicrobial defenses in insects started out of curiosity. We realized that insects in our breeding colonies of grasshoppers were very resistant to infections, and we didn't understand the mechanisms of this resistance," he said. At that time institutional funding didn't depend on relevance or potential applications. "Scientific curiosity was accepted as a reason to undertake our research. This was exactly 50 years ago; I am afraid that things would be much more difficult today," he added.

As much as the 32 to 64 gigabytes of internal storage in the standard HTC One might be enough for most users in the West, different markets sometimes call for different features. And a feature difference is exactly what we're seeing in the first leaked shots of what seems to be a Chinese HTC One variant going by the model name 802w. That's right -- there's dual SIM support, in addition to a microSD slot.

Spotted by Engadget Chinese over on Sina Weibo, the 802w hides all extra stuff behind a removable back cover. That's something not found on the European and American HTC One models, which pack everything into a sealed unibody enclosure with only a pop-out microSIM tray along the left edge. The Chinese handset, said to be headed to China Unicom, has one SIM slot for WCDMA/GSM (data and voice) and another for GSM (voice only), a trait common to many dual-SIM devices.

Even if it's a legit HTC-made product and not a knock-off, we wouldn't hold our breath for HTC One variants like this outside of the Far East. What's more, the prospect of using such a device without Google services might not make the 802w a particularly attractive prospect for import. But at least we'll always be able to fawn over the picture above.

(AP) ? The Senate gun control debate on the near horizon, a National Rifle Association-sponsored report on Tuesday proposed a program for schools to train selected staffers as armed security officers. The former Republican congressman who headed the study suggested at least one protector with firearms for every school, saying it would speed responses to attacks.

The report's release served as the gun-rights group's answer to improving school safety after the gruesome December slayings of 20 first-graders and six adults at a Newtown, Conn., elementary school. And it showed the organization giving little ground in its fight with President Barack Obama over curbing firearms.

Obama's chief proposals include broader background checks for gun buyers and bans on assault weapons and high-capacity ammunition magazines ? both of which the NRA opposes.

The study ? unveiled at a news conference watched over by several burly, NRA-provided guards ? made eight recommendations, including easing state laws that might bar a trained school staff member from carrying firearms and improving school coordination with law enforcement agencies. But drawing the most attention was its suggested 40- to 60-hour training for school employees who pass background checks to also provide armed protection while at work.

"The presence of an armed security personnel in a school adds a layer of security and diminishes the response time that is beneficial to the overall security," said Asa Hutchinson, a GOP former congressman from Arkansas who directed the study.

Asked whether every school would be better off with an armed security officer, Hutchinson replied, "Yes," but acknowledged the decision would be made locally.

It is unusual for guards to provide security at events that lack a major public figure at the National Press Club, which houses offices for many news organizations. NRA spokesman Andrew Arulanandam said he did not know whether the guards were armed, and several guards declined to say if they were.

Hutchinson said school security could be provided by trained staff members or school resource officers ? police officers assigned to schools that some districts already have.

Dan Domenech, executive director of the American Association of School Administrators, said while a trained law enforcement officer with a gun would be valuable, his group opposes arming "a teacher or an employee who simply has taken a course and now has the ability to carry a weapon."

The Brady Campaign, a leading gun-control group, accused the NRA of "missing the point" by ignoring the need for expanded background checks and other measures the Senate is considering. It said people want "a comprehensive solution that not only addresses tragic school shootings, but also helps prevent the thousands of senseless gun deaths each year."

Also denouncing the recommendations was Randi Weingarten, president of the American Federation of Teachers, which represents 1.5 million teachers and other workers. She called it a "cruel hoax that will fail to keep our children and schools safe" while helping only gun manufacturers.

The NRA released its report as congressional momentum seems to have stalled for any sweeping steps to curb firearms violence.

Top Senate Democrats have little hope for a proposed ban on assault weapons, and the prospects for barring large-capacity magazines also seem difficult. Key senators remain short of a bipartisan compromise on requiring gun transactions between private individuals to undergo federal background checks, which currently apply only to sales handled by licensed gun dealers. The Senate plans to begin debating gun legislation next week.

White House spokesman Jay Carney said administration officials were seeking middle ground and emphasized background checks, widely seen by gun control advocates as the most effective step available.

"We are working with lawmakers of both parties, and trying to achieve a compromise that can make this happen. Especially when it comes to the background checks," Carney told reporters.

The spokesman commented as a White House official revealed that the president plans a trip next week to Connecticut, scene of the horrific elementary school shootings that spurred the new push for gun control legislation. Obama wants to use the trip to build pressure on Congress to pass legislation.

Obama also plans to focus on firearms curbs in a trip Wednesday to Denver, not far from last summer's mass shooting in a movie theater in Aurora, Colo.

The 225-page study cost the NRA more than $1 million, Hutchinson said. The task force included several former top officials of federal law enforcement and security agencies, including the Secret Service and Homeland Security Department.

Hutchinson acknowledged that the study omitted an earlier NRA recommendation that retired police officers and other volunteers be armed to provide school safety. He said the idea encountered "great reluctance" from school superintendents.

Hutchinson said the NRA did not interfere with his task force's work. In a written statement, the NRA said the report "will go a long way to making America's schools safer."

Hutchinson also called "totally inadequate" a gun control measure working through the Connecticut legislature that includes a tightening of the state's assault weapons ban. The measure wouldn't prevent an attacker with a handgun or other firearms from attacking a school, he said.

Debbie Leidlein, chairwoman of the Newtown Board of Education, said having trained staff members carry weapons "can become a dangerous situation to have any individuals outside of those who have police training to be carrying weapons around children."

But the proposal won support from Mark Mattioli, whose 6-year-old son James was killed at Newtown and who attended the NRA news conference.

"These are recommendations for solutions, real solutions that will make our kids safer," Mattioli said.

Saudi court said to order criminal to be surgically paralyzed

DUBAI (Reuters) - Amnesty International has condemned a reported Saudi Arabian court ruling that a young man should be paralyzed as punishment for a crime he committed 10 years ago which resulted in the victim being confined to a wheelchair.

The London-based human rights group said Ali al-Khawaher, 24, was reported to have spent 10 years in jail waiting to be paralyzed surgically unless his family pays one million Saudi riyals ($270,000) to the victim.

The Saudi Gazette newspaper reported last week that Khawaher had stabbed a childhood friend in the spine during a dispute a decade ago, paralyzing him from the waist down.

Saudi Arabia applies Islamic sharia law, which allows eye-for-an-eye punishment for crimes but allows victims to pardon convicts in exchange for so-called blood money.

"Paralyzing someone as punishment for a crime would be torture," Ann Harrison, Amnesty's Middle East and North Africa Deputy Director, said in a statement late on Tuesday.

"That such a punishment might be implemented is utterly shocking, even in a context where flogging is frequently imposed as a punishment for some offences, as happens in Saudi Arabia," she added.

A government-approved Saudi human rights group did not respond to requests for comment.

The Arabic-language al-Hayat daily quoted Khawaher's 60-year-old mother as saying her son was a juvenile aged 14 at the time of the offence. She said the victim had demanded 2 million riyals to pardon her son and later reduced this to 1 million. "But we don't have even a tenth of this sum," she said.

Al-Hayat said an unnamed philanthropist was trying to raise funds to pay the blood money, but it was not clear how much time remained before Khawaher's sentence was to be carried out.

Amnesty said the case demonstrated the need for Saudi Arabia to review its laws to "start respecting their international obligations and remove these terrible punishments from the law".

Saudi judges have in the past ordered sharia punishments that include tooth extraction, flogging, eye gouging and - in murder cases - death.

Today's leafy Google doodle celebrates what would have been the 366th birthday of Maria Sibylla Merian, a naturalist and scientific illustrator whose beautifully detailed drawings of butterfly metamorphosis were a key early contribution to entomology.

Merian was born in 1697 in Frankfurt in what is now Germany, the daughter of an engraver and the stepdaughter of a still-life painter. She was fascinated by caterpillars from childhood, collecting as many as she could find and documenting in pictures how they change into butterflies and moths. When she grew up she kept her own live specimens, illustrating each stage of their life cycle - egg, larva, pupa, adult - in her work.

For much of her life, she was perhaps best known for publishing books of botanical drawings. Although popular with a high-society readership, they were apparently largely ignored by the scientific establishment because they were not written in Latin.

Careful observation

But her careful observations led to new insights about insect reproduction and the relationship between caterpillars and the plants on which they feed and lay their eggs. Her simple observational approach might not sound remarkable by today's standards, but it was pioneering for the time.

As well as being a scientific innovator, Merian was an intrepid explorer. In 1699, she secured funding from the city of Amsterdam to mount a scientific expedition to Suriname ? fifty years before it was explored by "Linnaeus's Apostle", Daniel Rolander. Surrounded by the spectacular natural bounty of the South American forest, she gathered and drew even more butterfly and moth specimens, as well as other insects and many plants.

Merian's illustrations, alongside those of living artist Rosemarie Trockel, can be seen at the Serpentine Gallery in London until the end of this week.
